Vice captain Helen Nelson is selected at fly-half and Christine Belisle packs down at tighthead prop.
Prop Anne Young and hooker Elis Martin, who joined Lightning this summer, are among the replacements as Scotland up their preparations ahead of WXV 2 later this month.
Scotland last played Wales in the 2024 Guinness Six Nations in an historic seventh Test win in a row, ending a 20-year-long spell of defeats in Cardiff with an 18-20 victory.
