Day, who only toured with England Under-18 to Australia last summer, has been chosen by head coach Rob Hunter on the replacements' bench.
The former student of St Joseph's College in Ipswich is one of eight players who represented England Under-18 last season and Hunter is excited to see what his players have to offer.
He said: "We're excited about getting into the Six Nations, it's a new side and we are all looking forward to seeing them play. We have a good blend of experience and fresh faces.
"Competition for places is high and we are pleased with how the team has come together. There are no easy games in this tournament and starting off against Scotland in Glasgow will be tough."
Friday night's game will be streamed live on www.rfu.com
